Rare copy number variants have a prominent role in the aetiology of schizophrenia and other neuropsychiatric disorders. Substantial risk for schizophrenia is conferred by large CNVs at several loci, including microdeletions at 1q21.1, 3q29, 15q13.3 and 22q11.2 and microduplication at 16p11.2. However, these CNVs collectively account for a small fraction of cases, and the relevant genes and neurobiological mechanisms are not well understood. Here we performed a large two-stage genome-wide scan of rare CNVs and report the significant association of copy (...) number gains at chromosome 7q36.3 with schizophrenia. Microduplications with variable breakpoints occurred within a 362-kilobase region and were detected in 29 of 8,290 patients versus 2 of 7,431 controls in the combined sample. All duplications overlapped or were located within 89 kilobases upstream of the vasoactive intestinal peptide receptor gene VIPR2. VIPR2 transcription and cyclic-AMP signalling were significantly increased in cultured lymphocytes from patients with microduplications of 7q36.3. These findings implicate altered vasoactive intestinal peptide signalling in the pathogenesis of schizophrenia and indicate the VPAC2 receptor as a potential target for the development of new antipsychotic drugs. © 2011 Macmillan Publishers Limited. Art’s Way Out invites readers to examine how what we call “art” enters a conditioned relationship with the polity that leaves the occasion of education and the condition of culture in a quandary that is at best inept and at worse oppressive. How and why did the notion of philosophy as a system evolve in Germany at the turn of the 16th and 17th centuries? Otto Casmann’s Modesta Assertio provides new answers to this question. Casmann, Clemens Timpler’s predecessor as professor in Steinfurt refers to other ‘like-minded philosophers’ who believe that philosophy is a ‘structured system of the liberal arts’. Casmann himself states that philosophy is a ‘structured unity of erudite wisdom’. The text is part of the debate between Daniel Hoffmann and (...) the Reformed philosophers about the relation between philosophy and theology. It can be made plausible that Hoffmann himself was Casmann’s target. The paper shows that a ‘structured unity of erudite wisdom’ presupposes harmony between theological insights and the findings of philosophy. Our thesis is that there is no moral requirement to refrain from emitting reasonable amounts of greenhouse gases solely in order to enjoy oneself. Joyriding in a gas guzzler provides our paradigm example. We first distinguish this claim that there is no moral requirement to refrain from joyguzzling from other more radical claims. We then review several different proposed objections to our view. These include: the claim that joyguzzling exemplifies a vice, causes or contributes to harm, has negative expected value, (...) exceeds our fair share of global emissions, and undermines political duties. We show why none of these objections succeeds and conclude that no good reason has yet been proposed that shows why joyguzzling violates a moral requirement. (shrink)

Origin of the German Trauerspiel was Walter Benjamin's first full, historically oriented analysis of modernity. Readers of English know it as "The Origin of German Tragic Drama," but in fact the subject is something else--the play of mourning. Howard Eiland's completely new English translation, the first since 1977, is closer to the German text and more consistent with Benjamin's philosophical idiom. Focusing on the extravagant seventeenth-century theatrical genre of the trauerspiel, precursor of the opera, Benjamin identifies allegory as the (...) constitutive trope of the Baroque and of modernity itself. This paper is an attempt to connect the Brazilian Paulo Freire’s well known educational thinking with the “philosophy for children” movement. It considers the relationship between the creator of philosophy for children, Matthew Lipman and Freire through different attempts to establish a relationship between these two educators. The paper shows that the relationship between them is not as close as many supporters of P4C have claimed, especially in Latin America. It also considers the context of Educational Policies in our time (...) and why Freire’s understanding of the politics of education makes it impossible to be Freirean and at the same time be neutral or favorable to the actual status quo. Finally, after presenting Lipman’s understanding of the relationship between philosophy, education and democracy and their connection to capitalism, it proposes ways to begin the political path of philosophizing with children inspired by Paulo Freire’s educational thinking. As a result, a more politically committed path to doing philosophy with children is offered. (shrink)
